The DHEA ELISA kit is a solid-phase enzyme-linked immunosorbent assay (ELISA), based onthe principle of competitive binding. The microtiter wells are coated with a polyclonal (rabbit)antibody directed toward an antigenic site on the DHEA molecule. During the first incubation,endogenous DHEA of a sample competes with a DHEA-horseradish peroxidase conjugate forbinding to the coated antibody. After incubation, the unbound conjugate is washed off, and thesolid phase is incubated with substrate solution. The colorimetric reaction is stopped by additionof stop solution, and the optical density (OD) of the resulting yellow product is measured. Theintensity of color is inversely proportional to the concentration of the analyte in the sample. Astandard curve is constructed by plotting OD values against concentrations of standards, andconcentrations of unknown samples are determined using this standard curve.
